Are there any shoujo manga out there that features one of it's characters, preferably the heroine, that loses weight? I mean like a lot, more than 20 pounds. Comedy would be nice.

hmmm...i don't think there are alot, largely because shoujo tends to love the bishoujo-ish main character.
well there's bara no tame ni, which is about an ugly girl who tries to get pretty by losing weight. but it's not a main plot element.
and, well, i know it isn't manga, but there's a korean drama called '200 pound beauty' which is about a woman with a beautiful voice but large body who is the 'voice' for this beautiful lip-synching idol. So then she disappears off the face of the earth for a while, loses weight, has plastic surgery,and returns as a skinny, beautiful girl and tries to enter the entertainment business through her own merits. It's funny, there's weight loss and it's an awesome storyline.

Miracle Dieter Miyuki is a comedy shoujo and deals with weight loss some, but I don't think any of the characters ever lose a large amount of weight.

Otometeki Koi Kakumei Love Revo!! Reverse Harem. Has a 100kg girl trying to lose weight. Not sure if she gets there because I havn't read that far.

is an interesting manga that has some weight lose but so far we don't know how much that she has lost. She is trying to remake her image entirely, not just weight but clothes and makeup too. But interestingly enough in high school she was popular.

Diet. From obesity to bulimia (or is it anorexia ?). But it's not scanlated into english yet.
